Etiology: Finding the Cause

Etiology is the scientific study of how diseases originate and develop. Understanding the specific causes of glioblastoma is vital not only for making sense of the diagnosis but also for identifying the cellular targets needed to create more effective and personalized treatments.

Known Risk Factors
  • Age: Incident rates for glioblastoma increase as individuals age, with the highest concentration of cases developing in adults over 60.
  • Prior Radiation: A significant risk factor is previous exposure to ionizing radiation, often as a result of earlier medical radiation therapy to the brain or neck area.
  • Genetic Syndromes: While most cases are sporadic, rare inherited syndromes such as Turcot syndrome or Li-Fraumeni syndrome are known to increase the risk.
Current Unknowns

Despite decades of intense clinical research, the precise cause for most individual cases remains unknown. Scientists are currently investigating gaps in knowledge regarding the roles of environmental chemicals, occupational exposures, and lifestyle habits. To date, there is no high-quality evidence linking common factors like mobile phone usage, power lines, or specific diets to the development of this tumor.

Cellular Origins

Glioblastoma originates from astrocytes—cells that normally provide support for neurons—or from their precursor stem cells. The process begins at a fundamental biological level when these glial cells suffer cumulative genetic mutations that disrupt their internal growth controls. These damaged cells fail to undergo natural cell death, leading to rapid, invasive growth into the surrounding healthy brain tissue.
